No, tolerance for psychoactive drugs usually results from the brain's own compensation from the introduction of the drug. Yes the amount a person takes can greatly increase this but in general the brain reacts to alterations that shouldn't be there. For example if you're taking medication that decreases the activity within the brain (i.e. to combat stress or to calm you down) the brain instinctively increases the number of receptors for the activity being inhibited. As a result you require more and more higher doses of the medication to produce the decrease in activity. The psychiatrist prescribes drugs that work, it's just the human body is a very unpredictable thing. And as you can see whether or not someone desires to be cured has no effect on whether or not they become tolerant to the meds they're on. These things just happen and produce a vicious cycle.
There are usually only 2 reason why the dead walk in these movies: 1) a hazardous biological compound, and 2) Sometimes no apparent reason (as seen in some of the much older films. Number 1 is usually more common.
Living Dead series? Or Resident Evil. The former are slightly worse in comparison but funny worse.
Zombies can be a very flexible plot device.
